I read Looking To Do Your First Modification? by Green-Buckshot today, and searched for more info on barrel physics.

What I'm wondering is, if one disables the overpressure release valve (in a plunger blaster, not flywheel I assume), won't the tube generate a small vacuum when the dart tries to leave a sealed barrel?

I think you mean air restrictor. Plunger blasters don't have an ORPV, but air powered blasters (ones with an air tank) do.

There are no OPRVs in sprung plunger blasters. They are only used in air pump blasters to limit the pressure which can be pumped in to a tank.
There is no vacuum created in a barrel of the ideal length, only if it was far too long.
